How to use
Presence/Absence
Add reagent to a 100mL sample, mix well, and incubate at 36°C ± 1°C for 24 hours.
Read results:
Colorless = negative
Yellow = total coliforms
Yellow/fluorescent = E. coli
Quantification
Add reagent to a 100mL sample and mix well.
Pour the sample into Elite-Tray/51 (counts from 1-200) or Elite-Tray/97 (counts from 1-2,419).
Seal in Elite-Tray Sealer and place in 36°C ± 1°C incubator for 24 hours.
Read results:
Yellow wells = total coliforms
Yellow/fluorescent wells = E. coli
Count positive wells and refer to the MPN table
Principle
The Science
The HuanKai Enzyme Substrate Coliform Test is designed to simultaneously detect total coliforms and E. coli. Two nutrient indicators, ONPG and MUG, are the major sources of carbon in our HuanKai's reagent and can be metabolized by the coliform enzyme β-galactosidase and the E. coli enzyme β-glucuronidase, respectively. As coliforms grow, they use β-galactosidase to metabolize ONPG and change it from colorless to yellow. E. coli use β-glucuronidase to metabolize MUG and create fluorescence. Since most non-coliforms do not have these enzymes, they are unable to grow and interfere. The few non-coliforms that do have these enzymes are selectively suppressed by HuanKai's specifically formulated matrix.
