Introduction

Handy plate® Rapid Colony Count Test Plate is a pre-prepared, ready-to-use culture medium product containing a standard culture medium, cold water gel and a composite color indicator. This product can be used for rapid counting of the total number of colonies in food and beverages.

How to use
1. Sample preparation: 1) Liquid samples: Mix well for later use. 2) Beverage concentrate and solid beverage samples: Take 25mL (g) of sample and place it in a sterile homogenizing bag containing 225mL of sterile phosphate buffer or sterile physiological saline, and beat it with a slapping homogenizer for 1min~2min to make a 1:10 sample homogenate. 3) According to the estimation of sample contamination, select 1~3 sample homogenates of appropriate dilution (liquid samples can be original solutions) for filtration inspection, and the filtration volume of each sample is not less than 10mL. 4) If the beverage contains larger particles that may interfere with filtration, use a filtration homogenizing bag for coarse filtration and then filtration. 2. Filtration: 1) Use sterile tweezers to clamp the filter membrane and place it on the sterilized filter bed, with the grid facing up, and connect the sterilized filter cup to the filtration device. 2) Aseptically transfer no less than 10mL of sample liquid into the filter cup and filter; when all the sample liquid is filtered, add no less than 15mL of sterile saline to the filter cup to rinse and filter. 3) After all the liquid passes through the filter membrane, remove the filter cup and remove the filter membrane with sterile tweezers; gently lift the upper membrane of the test piece (pre-moisten the test piece with 1mL of saline for about 2min, and it is easy to stick to the membrane if the wetting time is too long), place the filter membrane grid face down on the test piece, cover it with the upper membrane, and press it gently with your hand to prevent bubbles from forming. 3. Cultivation: Place the test piece horizontally with the front side facing up at 36±1 and culture for 24h±2h. If necessary, the culture time can be appropriately extended. 4. Interpretation: Turn the front side of the test piece over when observing, and count all color colonies (blue-green, blue-black, red, etc.) on the back regardless of size and clarity. The appropriate counting range is 30 CFU~300 CFU. If the entire culture area is light green and/or light red, the bacterial concentration may be too high and the sample needs to be further diluted to obtain an accurate count. If you need to separate the colonies for further analysis, peel off the upper membrane and use the inoculation needle to pick out the colonies for use.
