Bhk HP002 Coliform Count Plate Mini Culture Media Handy Plate for The Determination of Coliforms in Foods & Beverages

Coliform Count Plate is used for coliform count for sample from food / berverage etc.

Handy plate™ coliform test plates are pre-prepared, ready-to-use media products containing standard culture media, cold hydrogel, and chromogenic indicators. This product can be used for the determination of coliforms in food and beverages.

Instructions 

1. Sample preparation: Take 25 g (mL) of the sample and put it into a sterile homogenizing cup or homogenizing bag containing 225 mL of phosphate buffer (or physiological saline). Mix with the homogenizer for 1 to 2 minutes to prepare 1 :10 sample homogenate, if necessary, use sterile 1 mol/L NaOH or 1mol/L HCl solution to adjust the pH of the sample homogenate to 6.8~7.2. Use a 1mL sterile pipette or pipette to absorb 1 mL of the 1:10 homogeneous solution, inject it into a test tube containing 9 mL of diluent, shake it to become a 1:100 sample homogenate, and so on to prepare a 10-fold serial dilution of the sample. Homogenize the solution, changing the pipette one at a time.
2. Inoculation: Based on the estimate of sample contamination, select 2 to 3 dilutions for testing. Place the E. coli/coliform test piece on a flat experimental surface, uncover the upper film, use a sterile pipette to absorb 1 mL of uniform sample solution and add dropwise to the center of the test piece, slowly cover the upper film to avoid the generation of air bubbles, and let it sit Allow 3 to 5 minutes for the sample solution to diffuse and re-form the gel.
3. Culture: Place the test piece horizontally with the front side up in the incubator, and culture it at 36ºC±1ºC for 18h~24h.
4. Interpretation: Count all yellow acid rings and red colonies with bubbles around them. The appropriate counting range is 15 CFU ~ 150 CFU. If the bacterial concentration is too high and the entire culture area turns yellow and cannot be counted, the sample needs to be further diluted to obtain accurate counts. If you need to isolate bacterial colonies for further analysis, peel off the upper membrane and use an inoculation needle to pick out the colonies.
