Introduction
This kit is only suitable for qualitative detection of fecal streptococci in water bodies. Other tests must be carried out in accordance with the national standard "GB8538-2016 National Food Safety Standard Drinking Natural Water Detection Method". Principle
This kit is based on loop-mediated isothermal amplification (LAMP) technology and is used for the rapid detection of Burkholderia gladioli in food. The results can be directly interpreted with the help of the color change of the chromogenic indicator. . Two pairs of special primers and Bst DNA polymerase with strand displacement activity are used to circulate a circular single-stranded structure at the primer junction at both ends of the template. The primers successfully combine with the template and perform strand displacement amplification reaction, achieving constant temperature Continuous and rapid amplification is carried out under certain conditions (60~65). When a color indicator is added to the reaction, the color difference between negative and positive results is significant, and the results can be interpreted directly through the color change.

How to use?
1. Sample pretreatment A. Preparation of sample enrichment broth template DNA: a) Take 25 g of the sample to be tested and place it in 225 mL PDB enrichment broth, and enrich the bacteria at 36 ±1 for 8~12 h. This step can also be performed according to the method established by the laboratory; b) Take 1 mL of the above enrichment broth to a 1.5 mL sterile centrifuge tube, centrifuge at 6000 r/min for 5 min, and completely remove the supernatant; c) Add 30 μL lysis buffer to fully suspend the sediment at the bottom of the tube, tap the tube wall to eliminate bubbles, and heat at 99 for 10 min. d) Centrifuge at 12000 r/min for 15 min. The supernatant is the sample DNA to be tested. Leave it on ice and centrifuge it again if it is not used for a long time. B. Preparation of template DNA of suspicious colonies: Pick up a half circle of suspicious colonies and put it into a sterile centrifuge tube containing 30 μL lysis solution. After fully suspending, follow the above steps c) and d). 2. Mix reagents and add samples A. Reconstitution of negative and positive controls: When used for the first time, add 80 μL of ultrapure water to the positive control and negative control freeze-dried powder, mix after dissolving, and store at -20 for later use. B. Reconstitution and sample addition of dry powder reagent: 1) Take out the freeze-dried powder reagent, cut n test reagent tubes as needed (n = number of samples to be tested + 1 tube of positive control + 1 tube of negative control), and add 20 μL of reconstitution solution to each tube; 2) Add 2.5 μL of color indicator to each of the n reaction tubes to which the reconstitution solution has been added; 3) Add 2.5 μL of negative control, sample DNA to be tested, and positive control to the n reaction tubes in turn, and cover the lid immediately after each addition. 3. Amplification reaction: Use an instrument that can provide constant temperature conditions (preferably with a hot cover function), set the temperature to 64°C, and place the test tube in a constant temperature reaction for 50 min. 4. Result interpretation: After the reaction is completed, place the sample on a white background for observation in an environment with sufficient natural light. The negative control test tube will be light orange, and the positive control test tube will be fluorescent green. The test sample reaction tube will be interpreted based on this. If any result of the positive or negative control reaction tube does not match the above situation, the test result is invalid and should be retested or contact product technical support.
