Principle:

Peptone and yeast extract powder provide nitrogen sources, vitamins, amino acids, and carbon sources.Sucrose serves as a fermentable carbohydrate.Bacteriostatic agents inhibit most non-Vibrio cholerae bacteria.Sodium chloride maintains osmotic balance.Agar acts as the solidifying agent.Chromogenic substrates react specifically with enzymes produced by Vibrio cholerae, hydrolyzing the substrate to release chromogens, resulting in green-blue colonies on a pale yellow background.

Peptone 15.0g Yeast extract powder 5.0g Sucrose 10.0g Sodium chloride 5.0g Bacteriostatic agents 5.0g Agar 13.0g Chromogenic mixture 3.1g pH 9.2±0.2 at 25°C

How to use
Weigh 56.1 g of the medium and add to 1000 mL of distilled or deionized water.Stir and heat to boiling until fully dissolved. Cool to approximately 50°C.Optional: To enhance inhibitory effects, add 1 vial of Vibrio cholerae Selective Supplement (SR0450) per 100 mL medium under aseptic conditions. Mix thoroughly.Pour into sterile Petri dishes and allow to solidify.
